Effect of gamma radiation on hydration and structure of DNA - caffeine complex

Abstract

Measurements of dielectric permittivity for solutions of DNA, caffeine, and complex of DNA-caffeine were carried out. It is shown that the process of complex formation is accompanied by changes in dielectric properties. The degree of hydration (number of water molecules per one ligand molecule or one nucleotide) for all investigated substances was calculated. It was established that this value is equal 3 for caffeine, 16 - for DNA, and 14 - for the DNA-caffeine complex. For all samples, the values of Debye relaxation wavelength were determined. The irradiation of the complex results in some increase in the amount of bound water in the solution. It is shown that this effect can be caused by the growth of caffeine hydration. Parallel electrophoresis measurements showed that the degree of polyregularity for the irradiated complex is almost the same as for the initial sample. The absence of radiation influence on the hydration and the degree of fragmentation of DNA specifies protective properties of caffeine display at a molecular level.
