Na+ and Cd2+ ion effect on stability of triple helix POLY I - POLY A - POLY I
Abstract
Effects of Na+ (0.1÷2 M) and Cd2+ (10-6÷10-3 M) ions on the stability and structure of the three-stranded A2I were studied. It was shown that A2I melting is the 3 —> 1 (A2I —> A+I+I) transition. In accordance with polyelectrolyte theories, the temperature of this transition is a linear function of logarithm of the Na+ ion content in solution. The linear density of a charge on the single-stranded poly I surface (ξ=1,9) and the cooperativity factor of the 3 —> 1 transition in A2I (σ~0,001) were calculated. In the presence of Cd2+ ions A2I melting retains all the peculiarities of the 3 —> 1 transition but at the cadmium concentration exceeding 2 · 10-4 M the A2I thermal stability decreases. This results from the cadmium interaction with hypoxanthine N7, that leads to a change in the A2I structure which nevertheless at room temperature remains as the three-stranded helix up to 5 · 10-4 M Cd2+.
