Influence of mineralization on hydration and structural state of collagen
Abstract
The aim of the present work is to assess the influence of mineralization by hydroxyapatite on the process of hydration and the conformational state of collagen. The study was performed on the basis of two experimental techniques, namely infrared spectroscopy, and quartz piezogravimetry with subsequent mathematical processing of hydration isotherms according to D Arsy and Watt's model of absorption. All study samples were prepared in vitro. Our results indicate that mineralization leads to partial screening of hydration centers of collagen from water molecules resulting in the incomplete formation of the external hydration shell and increase of binding energies of water molecules of the internal hydration shell. Confirmation of triple helix in mineralized collagen differs from native tropocollagen molecule.
