Binding of theophylline to calf thymus DNA in presence of competing ligands
Abstract
The binding of theophylline to calf thymus DNA in presence of competing dyes has been studied. The actinolite derivative ActII and ethidium bromide were chosen as competing ligands that absorb in the Vis region and interact with DNA by different types of binding. It was shown that theophylline interacts with these ligands. The constants of theophylline binding with ActII (943±124 М-1) and ethidium bromide (177±36 М-1) were found to be higher than those for caffeine-ligand complexes. The examination of competitive DNA-theophylline and -ligand binding revealed that theophylline competes with ActII and ethidium bromide mainly in the range of low DNA concentrations. In contrast, in the range of high DNA concentrations, theophylline competes with ActII and ethidium bromide for the intercalative binding sites. The binding constant (185±20 М-1) and stoichiometry of binding (n=1) were obtained by means of DALSCOMP and COMPHET optimization programs. The constants of theophylline-DNA and caffeine-DNA binding were found to have a similar value of 190±15 М-1. Theophylline is more active than caffeine as an acceptor of planar biologically active molecules in the solution. This fact should be taken into account during the application of these substances in medicine and biology.
